さまざまなイメージング手法を取り入れ，生体内の分子を可視化することにより，疾患の原因解明および治療につながる基礎研究に取り組んでいます．特に，脳内環境・神経シナプス伝達に関わる機構とその破綻により生じる疾患に注目し，基礎研究を通して一人でも多くの人が長く健康な脳を保ち，生き生きと活躍できる社会づくりに貢献したいと考えています．

Title:MMP-2欠損マウスの硬組織デグラドームを利用した石灰化制御基質タンパクの探索
Project summary:骨と象牙質の石灰化に際しMMP-2で分解される「石灰化制御基質タンパク」を探索することを本研究の目的とする。MMP-2デグラドーム（MMP-2酵素で分解されるタンパクとその分解産物のプロテオーム）をMMP-2欠損マウスと野生型マウスのプロテオームを比較検討することで明らかとする。この際、プロテオームは質量顕微鏡を用いて可視化し、MMP-2欠損で残存し野生型で消失するタンパクを「石灰化制御基質タンパク」として抽出する。また、骨と象牙質のミネラル結晶の特性を解析し、MMP-2欠損における石灰化に関連する表現型も併せて検討する。平成３１年度では、MMP-2欠損マウス（GelA KO）を理研バイオリソース研究センター（理研BRC）から導入し、本学の動物実験施設における検疫を経て、繁殖を開始した。Mmp2-/-のメスは正常な分娩が行えないことから、Mmp2+/-の掛け合わせにより繁殖した。理研BRCの情報に基づき、遺伝子型決定のためのPCR条件の検討を行った。その結果、野生型配列の検出にはGelA Forward（5’-GTGCTACTGCAGGATAAACTGATG-3’）とGelA Reverse（5’-CCGGGACAGGAACGTACTGGGTTC-3’）のプライマーを用い、94℃ 2分の熱変成の後、98℃ 10秒、55℃ 30秒、68℃ 60秒の3ステップを30サイクル繰り返し、さらに72℃ 10分の伸長反応を行う反応条件を設定した。ノックアウト配列の検出には、GelA ForwardおよびPGKF（5’-CCGGGACAGGAACGTACTGGGTTC-3’）のプライマーを用い、94℃ 2分の熱変成の後、98℃ 10秒、58℃ 30秒、68℃ 60秒の3ステップを30サイクル繰り返し、さらに72℃ 5分の伸長反応を行う条件とした。

Title:質量顕微鏡データによる脳内環境の解読に向けた数理基盤の構築
Project summary:2018年度では、これまでに行なった解析結果をまとめて国際会議（北米神経科学会年会）でポスター発表を行なった。具体的には、まず、野生型マウスおよび遺伝子改変を行なったノックアウトマウスから採取した４つの脳標本スライスに対して質量顕微鏡イメージングによって質量スペクトルを得た。次に、一標本あたり1.5万枚となる質量スペクトル画像列に対して、適当な前処理（スペクトル強度が低すぎるものをカットする等）を施して数千枚程度まで減らした上で、代表的な線形的な多変量解析手法である主成分分析とISOMAPや局所線形埋め込み（LLE）、t-分布確率近傍埋め込み（t-SNE）などの非線形の低次元可視化手法である多様体学習を対象である質量スペクトル画像列に適用した結果を比較した。t-分布確率近傍埋め込みにおいて画像間の距離をコサイン距離にとった場合、主成分分析の結果に比べて物質特性の違いを反映してより詳細なクラスタ構造を得ることができた。しかし、この方法では野生型とノックアウト型の間で分布構造に明確な違いは現れなかった。以上の結果を発表した。
また、昨年度から行なってきた正準相関分析を一般化した方法（「WT同士あるいはKO同士は相関を高く、WTとKOの間の相関は低く」という付加情報を重み係数に反映させた手法）によって野生型とノックアウト型を直接比較する方法を引き続き試みたが、昨年度と同様明確な差を抽出することはできなかった。

Title:リポクオリティのイメージング
Project summary:平成27年度は質量顕微鏡法によりリポクオリティの変化を解析した。静岡大学との共同研究で運動によって骨格筋においてリン脂質のリポクオリティが変化し、それにはPGC-1αが関与していることを発見した（Senoo N et al., 2015）。また、産婦人科との共同研究で胎盤血流障害におけるスフィンゴ脂質のリポクオリティの病的な変化を見出した（Yamazaki K et al., 2015）。外科との共同研究で腹部大動脈瘤におけるリポクオリティの動態解析を解析した（Tanaka et al., 2015; Tanaka et al., 2015）。東大等との共同研究で腸管で作用する脂質メディエーター17,18-EpETE動態の変化（Kunisawa et al., 2015）を明らかにした。加えて、本年度はリポクオリティの可視化に有用となる新規質量顕微鏡解析法を開発し、硫酸アンモニウム添加によるリン脂質の検出感度向上法を開発した（Sugiyama et al., 2015）。さらに線虫におけるリポクオリティ可視化に適した質量顕微鏡解析法を開発した（Hameed S et al., 2015）。
また、エンバクの青色光受容体タンパク質を用い青色光で細胞内脂質を操作する実験系の立ち上げを行った。遺伝子導入を行う細胞については、チャンバーシステムを用いて、神経細胞体と軸索との区画分けを可能とする初代培養系の立ち上げを行った。光操作を行う顕微鏡システムについても、高解像度での細胞内の局所的な青色光の照射、およびヘテロ二量体の結合、乖離をリアルタイムでの観察、記録を可能とするシステムの立ち上げを行った。

Title:Super-resolution imaging of synaptic proteins regulated with ubiquitin ligase SCRAPPER
Project summary:Dysfunction of the ubiquitination occurs in many neurodegenerative diseases, and neurotransmitter release abnormalities are also involved in many neurological diseases. We tried to understand integratively the regulation of cell and individual function by protein degradation through ubiquitin ligase SCRAPPER which we previously identified. In this study, we found that the amounts of some kinds of proteins were significantly altered in the knockout mouse brain. Moreover, it has become clear that behaviors of several neurotransmitters have changed in the multiple areas of the knockout mouse brain. These alterations may have deep relationship with the roles of SCRAPPER in neural tissue such as neurotransmitter release control, neural plasticity, and fear / anxiety-like behavior.

Title:Visualisation of the marker for oligodendrocytes by imaging mass spectrometry
Project summary:Sulfatide species comprise the major glycosphingolipid components of myelin. Although various sulfatides contain different fatty acids, it is unclear how these sulfatides affect myelination. The O4 antibody reaction with sulfatide has been widely used as a useful marker for oligodendrocytes/myelin. However, sulfatide synthesis during the pro-oligodendroblast stage has not been examined. Here, we identified a sulfatides from the pro-oligodendroblast to myelination stage by imaging mass spectrometry. The results demonstrated short-chain sulfatides with 16 carbon fatty acids (C16) and C18 existed in the area, where pro-oligodendroblasts initially appear. C22 sulfatide became predominant later in oligodendrocyte development and the longer C24 sulfatide was predominant in the adult brain. These findings indicated that O4 recognized sulfatides with short chain fatty acids in pro-oligodendroblasts and individual sulfatide species may have unique roles in oligodendrogenesis and myelination.
