KAMADA YUKA
私は膜タンパク質 CFTR の変異体を主なモデル基質として、異常膜タンパク質がどのように分解されるのかを研究しています。野生型 CFTR は小胞体で正常にフォールディングされ、適切な発現場所へと輸送されます。しかし変異型 CFTR (例えば ∆F508-CFTR) は適切にフォールディングされず分解されてしまうため、適切な場所で発現できず、嚢胞性線維症 (CF) を引き起こします。この過程において、ユビキチン (Ub) 化酵素 (E3) が異常タンパク質を認識し Ub 化することで分解を促進します。
E3 が異常タンパク質を認識するメカニズムを明らかにすることは、膜タンパク質の変異が引き起こす疾患の治療法開発に貢献できると期待されます。
